Colony PCR

  1. Place 50 ul of ddH2O in a 1.5 ml tube.
  2. Scrape a decent-sized colony from a plate and dissolve in the water.
  3. Heat at 95-98 °C for 5-10 min.
  4. Centrifuge at 12000 x g for 1 min.
  5. Use 10 ul of the supernatant as a template for the following PCR:
Ingredient Volume
template 5 ul
oligo 1 1.25 ul
oligo 2 1.25 ul
Q5 2X MM 12.5 ul
water 5 ul
