Analyze intact mass on Compass

  • in compass, on your TIC/BPC, right click and drag to select small range
  • This will display in spectrum view. Look for an envelope. Confirm that noise is low-ish
  • right click copy to compound spectra
  • you can use charge state rule (on top toolbar) to check that you're looking at the right protein (approximate mass)
  • Deconvolute->parameters
  • -max entropy mode
  • -change masses to narrow range around your protein (+-1kD)
  • -Instrument resolving power: 50,000
  • -resolution: High
  • -Apply, OK
  • Deconvolute->Mass spectra
  • Wait for green bar
  • expand the compound entry to reveal deconvoluted entry below
  • zoom out of compound spectra to see intact mass envelope
  • celebrate a job well done
