As title - Two-Hand changes a weapon's damage die to the one listed in the trait, which Deadly Simplicity does not buff to the next die size up.

More specifically, the two hand trait is a change, but its a change to a higher value therefore informally an "increase". You cannot increase damage dice more than once.
