Hi
I read the paper published and it is stated that the primer is designed to anneal between 65 - 68 but somehow I checked the config.py file stated that the temperature was set between 59.5 to 62.5 I never change anything after I downloaded the program. So I wonder if this default actually outputs primer with annealing temperature between 65-68 or I will have to change them in config.py?

That's because Q5 polymerase annealing temperature is significantly higher than other polymerases; https://tmcalculator.neb.com/#!/main
